位於物理系館南棟，「原理化學實驗室」是創校以來最古老的建築物，從廊道觀之，可見建築設計所具有的日式風情。考量到臺灣潮濕炎熱的氣候，牆面除了有整排的上下推窗，可供室內通風外，抬高的樓板亦可用於防潮，而建物前後的水池則可在炎炎夏日達到調節溫度的作用。圖書課是創校後的第二棟建築物，此閱覽空間整體呈現內凹作法，平屋頂的半戶外廊道為其特色，造成了相對低矮的視覺效果。內凹的空間容納了一座半月形水池，許是與儒家「思樂泮水」的寓意有關，欲藉此勉勵莘莘學子奮發學習。

The oldest building of NCKU, located on the Cheng Kung campus, is the Physical Chemistry Laboratory (currently known as the Frontal Building of the Department of Physics). This simple one-story building was completed in 1930 and has served various purposes, such as being the joint laboratory of all the departments, the infirmary, the lecture hall, the teaching staff office, and the storage. It is a witness to many of the school's earliest ceremonies, including the very first student admission ceremony.
